Experimental Studies on Fibrin Thrombus Formation in the Hepatic Sinusoids Induced by Oral Contraceptives

Abstract
The potential dangers involved in the use of oral contraceptive have been of concern for years and are still a vexing issue. The side actions were recently reported by many authors and much depends upon the amount taken, the duration of use, the potency of the preparation, and the length of the follow up. Attention was paid recently for the increased incidence of venous thrombosis due to adminstration of oral contraceptives. This study was carried out to investigate the influence of long duration treatment of oral contraceptive (Ovral) in rats. Each plasma protein fraction levels were estimated. And both light and EM observation were done. Also observation were carried out in estrogen and progesterone treated groups. The results were as follows: 1. In blood plasma, ¥á©û and ¥á©ü globulin showed increased levels in animal groups treated with Ovral or estrogen. 2. Fibrin thrombus formation in the hepatic sinusoid was seen in animal groups treated with Ovral or estrogen. 3. Liver showed regressive changes in Ovral and estrogen treated groups, while progesterone treated group showed mild changes compared with former two groups. 4. Adenomatous hyperplasia of the endometrial gland was induced in both estrogen and progesterone treated animal groups. With the above results, it was concluded that globulin levels in the blood plasma and fibrin thrombus formation in the hepatic sinusoid may have a close relationship in animmals treated with Ovral and estrogen.
